Forest restoration is mainly based on plant-soil relationships and plant species with economic potential, but those between insects and other arthropods are also important to this reestablishment. The objective was to evaluate, during 24 months, the relationships between tending ants, Hemiptera phytophagous, predators and their distribution pattern (aggregated, random or uniform). The arthropods were sampled, stored and identified and their relationships and distribution patterns calculated with the BioDiversity-Pro software. The number of tending ants and phytophagous Hemiptera, Brachymyrmex sp. and Aethalion reticulatum, Cephalotes and Aleyrodidae were positively correlated. Tending ants were negatively correlated with Sternorrhyncha predators on A. auriculiformis saplings. The distribution of arthropods was aggregated, except for Teudis sp. and Cephalocoema sp., with a random pattern. The herbivores Stereoma anchoralis, Aethalion reticulatum and Tetragonisca angustula and the predators Brachymyrmex sp. and Dolichopodidae were the most abundant arthropods. The relationships between the arthropods studied on A. auriculiformis indicate that this plant, even introduced, is suitable for programs to recover degraded areas in the savannah.

Canine parvovirus 2 (CPV-2), a highly contagious virus, affects dogs worldwide. Infected animals present severe and acute gastroenteritis which may culminate in death. CPV-2 VP2 protein is responsible for important biological functions related to virus-host interactions. Herein we obtained VP2 full-length gene sequences from Brazilian dogs with bloody diarrhea (n=15) and vaccine strains (n=7) produced by seven different laboratories and marketed in Brazil. All wild sequences and one vaccine strain were classified as CPV-2b and six vaccines were the classic CVP-2. Mutations in VP2 protein from vaccine and wild strains obtained in Brazil and worldwide were analyzed (n=906). Amino acid sequences from vaccine strains remarkably diverge from each other, even that classic CPV-2. Phylogenetic analysis based on VP2 gene and conducted with sequences displaying mutations in epitope regions previously described shows that vaccine strains are distantly related from the wide range of wild CPV-2. The impact of amino acid mutations over VP2 protein structure shows that vaccine and wild strains obtained in this study diverge in loop 3, an epitope region that plays a role in the CPV-2 host range. This is the first analysis of CPV-2 VP2 from commercial vaccine strains in Brazil and wild ones from Minas Gerais State, Brazil, and the first detailed attempt to vaccinal VP2 molecular and structural analyses.

Surgical site infections (SSIs) and antimicrobial resistance among pathogens causing SSI are a growing concern in veterinary hospitals. One major reason, the widespread use of antimicrobials, has led to increased incidence of SSIs. This study identified bacteria and resistance profiles to antimicrobials in the SSI cases diagnosed at the Surgical Clinic of Small Animals in the Veterinary Hospital, Federal University of Viçosa, Brazil. The main genus identified was Staphylococcus, followed by Escherichia, Enterococcus, Bacillus, Shigella, Citrobacter, Proteus, Morganella, Serratia, Enterobacter, Pseudomonas and Klebsiella were also found, but in small number. The results indicated the predominance of Gram-negative bacteria among the collected samples. Most of isolates identified were resistant to more than one of the following antimicrobials: ampicillin, tetracycline, enrofloxacin, amoxicillin/clavulanic acid and cephalotin. Of the 17 Staphylococcus sp. isolates, two (11.8%) were methicillin-resistant Staphylococcus aureus (MRSA) and 11 (64.7%) of them were methicillin-resistant Staphylococcus pseudintermedius (MRSP). There were bacterial genera identified with resistance to all tested antimicrobials in different proportions. This should alert veterinary hospitals to the emergence of multidrug-resistant bacteria and to the requirement for the revision of surgical protocols with regard to antimicrobial prophylaxis and therapy.(AU)


As infecções em sítio cirúrgico (ISCs) e a resistência bacteriana entre os patógenos relacionados constituem uma preocupação crescente nos hospitais veterinários. O aumento na incidência de ISCs possui forte relação com o uso amplo e disseminado de antibióticos. O presente estudo identificou bactérias e perfis de resistência a antibióticos nos casos de ISCs diagnosticados na Clínica Cirúrgica de Pequenos Animais do Hospital Veterinário da Universidade Federal de Viçosa, Brasil. O principal gênero identificado foi Staphylococcus, seguido pelos gêneros Escherichia, Enterococcus, Bacillus, Shigella, Citrobacter, Proteus, Morganella, Serratia, Enterobacter, Pseudomonas e Klebsiella, porém, em menor quantidade. Os resultados demonstraram a predominância de bactérias Gram-negativas entre as amostras coletadas. A maioria dos isolados identificados eram resistentes a um ou a mais de um dos seguintes antibióticos: ampicilina, tetraciclina, enrofloxacina, amoxicilina/ácido clavulânico e cefalotina. Entre os 17 isolados de Staphylococcus sp., dois (11,8%) eram Staphylococcus aureus resistentes à meticilina (SARM) e 11 (64,7%) eram Staphylococcus pseudintermedius resistentes à meticilina (SPRM). Houve identificação de gêneros bacterianos com diferentes proporções de resistência para todos os antibióticos avaliados. Esses achados devem alertar os hospitais veterinários para a emergência de bactérias multirresistentes e para a necessidade de revisar a profilaxia e a terapia antimicrobiana referente aos protocolos cirúrgicos.(AU)

This study was performed to evaluate the compressive mechanical strength of rigid internal fixation (RIF) using 1.5-mm L-shaped plates fixed with monocortical screws in sagittal split osteotomy (SSO). Thirty synthetic hemimandibles, which had all undergone a 5-mm advancement, were divided into three groups: three 12-mm bicortical titanium screws were placed in an inverted L pattern in group A; one straight 2.0-mm system spaced titanium plate fixed with four 5-mm monocortical screws was used in group B; two 1.5-mm system L-shaped titanium plates, each fixed with four 5-mm monocortical screws, were used in group C. The models were subjected to compressive and progressive mechanical tests with forces applied in the area between the second premolar and first molar to verify resistance in Newtons (N). A displacement speed of 1mm/min was applied, with a maximum 10mm displacement of the distal segment or until disruption of the fixation. The deformity and/or eventual rupture of the plates were evaluated, and consequently their technical stability was determined. The results showed that the modified fixation technique tested in this study on synthetic mandibles resulted in adequate stability and superior mechanical behaviour compared to simulated osteosynthesis with the use of a straight 2.0-mm titanium plate.

O vírus da bronquite infecciosa (IBV) é um importante patógeno respiratório presente na avicultura comercial e tem provocado grandes perdas econômicas em todo o mundo. A vacinação é realizada pela indústria produtora de aves, mas continuam surgindo novos sorotipos e variações antigênicas, dificultando o controle de IBV. Nós realizamos uma caracterização molecular de uma cepa de IBV obtida diretamente de tecidos e comparamos com a mesma cepa que havia sido passada três vezes em ovo embrionado. Nós mostramos uma variação significante na sequência viral depois de ter sido isolada em ovo embrionado.(AU)

Visceral Leishmaniasis (VL) represents an important global health problem in several warm countries around the world. The main targets in this study are the two nucleoside triphosphate diphosphohydrolases (NTPDases) from Leishmania infantum chagasi that are the main etiologic agent of VL in the New World. These enzymes, called LicNTPDase1 and -2, are homologous to members 5 and 6 of the mammalian E-NTPDase/CD39 superfamily of enzymes. These enzymes hydrolyze nucleotides and accordingly can participate in the purine salvage pathways and in the modulation of purinergic signaling through the extracellular nucleotide-dependent host immune responses. They can therefore affect adhesion and infection of host cells and the parasite virulence. To further characterize these enzymes, in this work, we expressed LicNTPDase1 and -2 in the classical bacterial system Escherichia coli and mammalian cell system COS-7 cells. Our data demonstrate that changes in refolding after expression in bacteria can increase the activity of recombinant (r) rLicNTPDase2 up to 20 times but has no significant effect on rLicNTPDase1. Meanwhile, the expression in COS-7 led to a significant increase in activity for rLicNTPDase1.

Brazilian pig population is made up of several naturalized breeds; among them the Piau breed is known for its rusticity and large fat stores. The naturalized breeds, in comparison with commercial ones, may have an increased resistance to diseases circulating in their territory. Thus, this study aimed to verify if there are differences between the serologic profile against Porcine circovirus 2 (PCV2) of Piau pigs and that of a commercial breed from a farm naturally infected by PCV2. The serum viral load was measured by qPCR, and levels of anti-PCV2 antibodies were measured by ELISA. The results showed that the serum viral load was similar across all animals. However, Piau piglets showed higher levels of antibodies compared to commercial piglets (P= 0.05), while sows of the commercial breed showed higher levels than the Piau breed (P< 0.01). There was not a statistical difference between pigs of different production stages in the seroprevalence of PCV2 or the blood viral load. This work demonstrates that, with regard to a natural PCV2 infection, the Piau breed has a different humoral immune response compared to the response developed by the commercial pigs. The results support the importance of conservation of native breeds.(AU)


O rebanho de suínos brasileiro é constituído por diversas raças naturalizadas, entre elas a raça Piau, que é conhecida por sua rusticidade e pela grande deposição de toucinho. As raças naturalizadas, em comparação com as linhagens comerciais, podem ter uma maior resistência a doenças que circulam em seu território. Dessa forma, o presente estudo teve como objetivo verificar se existem diferenças no perfil sorológico contra o Porcine circovirus 2 (PVC2) entre suínos da raça Piau e de uma linhagem comercial de uma granja naturalmente infectada pelo PCV2. Foram realizadas mensurações da carga viral sérica por qPCR e dos níveis de anticorpos anti-PCV2 por meio da técnica de ELISA. Os resultados mostraram que a carga viral sérica se manteve homogênea em todos os animais e que os leitões da raça Piau apresentaram níveis de anticorpos superiores em comparação com os leitões da linhagem comercial (P=0,05), enquanto as porcas de linhagem comercial apresentaram níveis superiores aos da raça Piau (P<0,01). Este trabalho fornece indícios de que a raça Piau apresenta uma resposta imune humoral distinta diante de uma infecção natural pelo PCV2, quando comparada com a resposta desenvolvida pela linhagem comercial. Os resultados obtidos reforçam a importância da conservação das raças nativas.(AU)

Fish germ cell transplantation presents several important potential applications for aquaculture, including the preservation of germplasm from endangered fish species with high genetic and commercial values. Using this technique in studies developed in our laboratory with adult male Nile tilapias (Oreochromis niloticus), all the necessary procedures were successfully established, allowing the production of functional sperm and healthy progeny approximately 2months after allogeneic transplantation. In the present study, we evaluated the viability of the adult Nile tilapia testis to generate sperm after xenogeneic transplant of germ cells from sexually mature Jundia catfish (Rhamdia quelen) that belong to a different taxonomic order. Therefore, in order to investigate at different time-periods post-transplantation, the presence and development of donor PKH26 labeled catfish germ cells were followed in the tilapia seminiferous tubules. From 7 to 20days post-transplantation, only PKH26 labeled spermatogonia were observed, whereas spermatocytes at different stages of development were found at 70days. Germ cell transplantation success and progression of spermatogenesis were indicated by the presence of labeled PKH26 spermatids and sperm on days 90 and 120 post-transplantation, respectively. Confirming the presence of the catfish genetic material in the tilapia testis, all recipient tilapias evaluated (n=8) showed the genetic markers evaluated. Therefore, we demonstrated for the first time that the adult Nile tilapia testis offers the functional conditions for development of spermatogenesis with sperm production from a fish species belonging to a different order, which provides an important new venue for aquaculture advancement.

RESUMO A espécie Mikania glomerata Sprengel, popularmente conhecida no Brasil como guaco, é amplamente utilizada como expectorante para tratar doenças respiratórias e tem a sua atividade farmacológica atribuída principalmente a cumarina. Os resultados mostraram que o método apresenta linearidade de 0,05 a 0,8 mg mL-1. Ele foi considerado seletivo, exato e preciso. A proposta de um método rápido para determinação de cumarina em extratos de guaco torna-se interessante para a rotina de controle de qualidade industrial, visando à obtenção de medicamentos fitoterápicos padronizados.


ABSTRACT The species Mikania glomerata Sprengel, popularly known in Brazil as “guaco”, is widely used as an expectorant to treat respiratory diseases. Its pharmacological activity is mainly attributed to coumarin. The results showed that the method for determining coumarin presented linearity from 0.05 to 0.8 mg mL-1. It was considered selective, accurate, and precise according to the specific resolution from ANVISA, the Brazilian regulatory agency. The proposal of a rapid method for the determining coumarin in extracts of guaco is interesting for routine industrial quality control in order to obtain standardized, efficient, and safe phytotherapic medicines.

Avaliou-se o efeito do flavonoide quercetina na replicação do parvovírus canino in vitro por meio do ensaio de determinação da atividade virucida (ensaio 1), ensaio de determinação da atividade sobre a célula (ensaio 2) e ensaio de tempo de adição das drogas em diferentes etapas do ciclo replicativo viral (ensaio 3). A quercetina apresentou significante atividade antiviral, com valores máximos de redução do título viral de 96,3% no ensaio 1, 90% no ensaio 2 e 90% no ensaio 3. Os efeitos mais expressivos ocorreram nas etapas de adsorção e penetração viral. Os resultados deste trabalho sugerem a importância da quercetina para a medicina veterinária.


The in vitro effect of the flavonoid quercetin against canine parvovirus was evaluated. The antiviral activity of quercetin was evaluated by determining the virucidal activity (assay 1), determining the activity on the cell (assay 2) and using the time of addition assay to test the inhibition of the viral replication cycle (assay 3). Quercetin showed a significant antiviral activity, with maximum viral titer reduction of 96.3% in assay 1, 90% in assay 2 and 90% in assay 3. The most expressive effects occurred in the stages of viral adsorption and penetration. The results show the importance of quercetin for veterinary medicine.

We previously demonstrated that reactive oxygen species (ROS) could be involved in ultraviolet-C (UVC)-induced DNA damage in Escherichia coli cells. In the present study, we evaluated the involvement of the GO system proteins in the repair of the 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxoG, GO) lesion, which is ROS-induced oxidative damage. We first found that the mutant strain Δfur, which produces an accumulation of iron, and the cells treated with 2,2'-dipyridyl, a iron chelator, were both as resistant to UVC-induced lethality as the wild strain. The 8-oxoG could be mediated by singlet oxygen ((1)O(2)). The Fpg protein repaired this lesion when it was linked to C (cytosine), whereas the MutY protein repaired 8-oxoG when it was linked to A (adenine). The survival assay showed that the Fpg protein, but not the MutY protein, was important to UVC-induced lethality and interacted with the UvrA protein, a nucleotide excision repair (NER) protein involved in UVC repair. The GC-TA reversion assay in the mutant strains from the '8-oxoG-repair' GO system showed that UVC-induced mutagenesis in the fpg mutants, but not in the MutY strain. The transformation assay demonstrated that the Fpg protein is important in UVC repair. These results suggest that UVC could also cause indirect ROS-mediated DNA damage and the Fpg protein plays a predominant role in repairing this indirect damage.

Plantas medicinais são utilizadas mundialmente como uma das principais formas de cuidado primário de saúde. No entanto, a literatura indica que muitas espécies podem apresentar composição química variável, toxicidade ou difícil identificação. O objetivo do presente estudo foi obter critérios úteis para o controle de qualidade farmacognóstico das principais espécies vegetais de uso popular no estado do Rio Grande do Norte, utilizando metodologias de análise botânica (morfodiagnose macro e microscópica) e química (triagem fitoquímica e cromatografia em camada delgada), evitando assim adulterações ou uso inadequado dessas plantas medicinais no estado. No total, sete espécies foram analisadas Acmella oleracea, Chenopodium ambrosioides, Lippia alba, Mentha piperita, Ocimum gratissimum, Peumus boldus e Rosmarinus officinalis. Diversos marcadores botânicos e fitoquímicos foram identificados, contribuindo dessa forma para a correta identificação destas espécies de plantas medicinais importantes no estado do Rio Grande do Norte.


Medicinal plants are worldwide used as one of the main forms of primary healthcare. However, the literature indicates that many species may have variable chemical composition, toxicity, or even difficult identification. The aim of this study was to obtain useful criteria for pharmacognostic quality control of the main plant species of popular use in the state of Rio Grande do Norte, using methods of botanical (macro and microscopic morphodiagnosis) and chemical (phytochemical screening and thin-layer chromatography) analysis, thus preventing adulteration or inappropriate use of these medicinal plants in the state. In total, seven species were analyzed Acmella oleracea, Chenopodium ambrosioides, Lippia alba, Mentha piperita, Ocimum gratissimum, Peumus boldus and Rosmarinus officinalis. Several botanical and phytochemical markers were identified, thereby contributing to the correct identification of these important medicinal plant species in the state of Rio Grande do Norte.

We previously demonstrated that reactive oxygen species (ROS) could be involved in the DNA damage induced by ultraviolet-C (UVC). In this study, we evaluated singlet oxygen ((1)O(2)) involvement in UVC-induced mutagenesis in Escherichia coli cells. First, we found that treatment with sodium azide, an (1)O(2) chelator, protected cells against UVC-induced lethality. The survival assay showed that the fpg mutant was more resistant to UVC lethality than the wild-type strain. The rifampicin mutagenesis assay showed that UVC mutagenesis was inhibited five times more in cells treated with sodium azide, and stimulated 20% more fpg mutant. These results suggest that (1)O(2) plays a predominant role in UVC-induced mutagenesis. (1)O(2) generates a specific mutagenic lesion, 8-oxoG, which is repaired by Fpg protein. This lesion was measured by GC-TA reversion in the CC104 strain, its fpg mutant (BH540), and both CC104 and BH540 transformed with the plasmid pFPG (overexpression of Fpg protein). This assay showed that mutagenesis was induced 2.5-fold in the GC-TA strain and 7-fold in the fpg mutant, while the fpg mutant transformed with pFPG was similar to GC-TA strain. This suggests that UVC can also cause ROS-mediated mutagenesis and that the Fpg protein may be involved in this repair.

Gymnema sylvestre extract (GSE) is a plant product widely used as an adjuvant in the treatment of diabetes mellitus and commercially available as a powder. Owing to its low flowability, the manufacturing of hard gelatin capsules containing GSE faces specific problems. The purpose of this study was to investigate the best excipient (starch, lactose or microcrystalline cellulose) for hard gelatin capsules containing GSE. The technological properties such us bulk density (ñâ); tapped density (ñt); inter-particle porosity (Ie); Carr index (CI); Hausner ratio (HR); loss on drying (%LOD) and particle size distribution (%Pf) of the various GSE mixtures were investigated with the aim of identifying the best excipient. The need for lubricants (talc/magnesium stearate) was also assessed. GSE was characterized as a fine powder with more than 50% of its particles between 0.149mm to 0.250mm; furthermore, CI=25.6%; RH=1.3 and Ie = 25.6% and, as expected with such properties, it showed poor flowability. All the excipients investigated were able to change the technological properties of GSE and the powder mixture containing microcrystalline cellulose gave the best results.


O extrato seco de Gymnema sylvestre (EGS) é um produto fitoterápico amplamente utilizado como adjuvante no tratamento da diabetes, sendo comercializado na forma de pó. O objetivo do presente estudo foi investigar a influência da adição de adjuvantes (amido, lactose ou celulose microcristalina) à formulação para a preparação de cápsulas gelatinosas duras contendo o EGS. As propriedades tecnológicas como densidade aparente (Da), densidade aparente de compactação (Dc), porosidade interparticulas (Ie), Índice de Carr (IC), Fator de Hausner (FH), perda por dessecação (PD%) e análise do tamanho de partículas (%Pf) das diferentes misturas preparadas foram investigadas com o objetivo de escolher o melhor excipiente. A necessidade de agentes lubrificantes (talco/estearato de magnésio) também foi avaliada. O EGS foi caracterizado como um pó fino, com mais de 50% do material particulado compreendido entre 0,149-0,250mm; IC=25,6%; FH=1,3 and Ie = 25,6%, o que justifica seu fluxo pobre. Todos os excipientes testados foram capazes de modificar as propriedades tecnológicas do EGS, sendo a mistura de pós que apresentou melhores resultados aquela obtida com a adição de celulose microcristalina.

The antiviral activity of quercetin, morin and trans-cinnamic acid was evaluated in vitro against equid herpesvirus 1 (EHV-1) by determining the virucidal activity and using the time of addition assay to test inhibition of the viral replication cycle. The cytotoxicity of each substance was assessed using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide]. Quercetin showed virucidal action and inhibition of the viral replication cycle at 0 and 1h. Morin showed potential virucidal and viral replication cycle inhibition at 0 h. Trans-cinnamic acid did not show virucidal activity but inhibited the viral replication cycle at -1 and 0 h. This study demonstrates the potential of these compounds as future antiviral candidates in relation to viruses of importance in veterinary medicine.

Porcine circovirus-2 (PCV-2) is the main agent related to post-weaning multisystemic wasting syndrome (PMWS) and it is also associated with other syndromes affecting pigs. Not all pigs infected with PCV-2 will develop PMWS and the incidence of PMWS is higher when coinfecting viral and bacterial pathogens are present. In this study, PCV-2 viral loads were evaluated in the tissues of animals with and without PMWS in order to investigate the relationship between viral load and microscopical lesions. Lymph nodes had the highest average viral load, but there was no significant difference between lesion severity and the viral load in these structures. There was no significant difference between the average viral load in inguinal lymph nodes of animals with and without PMWS. However, samples from pigs with PMWS had more severe lesions compared with samples from non-PMWS animals. These findings suggest that other infectious and non-infectious cofactors may be important in the pathogenesis of PMWS.

Triamcinolone (TRI), a drug widely used in the treatment of ocular inflammatory diseases, is practically insoluble in water, which limits its use in eye drops. Cyclodextrins (CDs) have been used to increase the solubility or dissolution rate of drugs. The purpose of the present study was to validate a UV-Vis spectrophotometric method for quantitative analysis of TRI in inclusion complexes with beta-cyclodextrin (B-CD) associated with triethanolamine (TEA) (ternary complex). The proposed analytical method was validated with respect to the parameters established by the Brazilian regulatory National Agency of Sanitary Monitoring ANVISA). The analytical measurements of absorbance were made at 242nm, at room temperature, in a 1-cm path-length cuvette. The precision and accuracy studies were performed at five concentration levels (4, 8, 12, 18 and 20µg.mL-1. The B-CD associated with TEA did not provoke any alteration in the photochemical behavior of TRI. The results for the measured analytical parameters showed the success of the method. The standard curve was linear (r2 > 0.999) in the concentration range from 2 to 24 µg.mL-1. The method achieved good precision levels in the inter-day (relative standard deviation-RSD <3.4%) and reproducibility (RSD <3.8%) tests. The accuracy was about 80% and the pH changes introduced in the robustness study did not reveal any relevant interference at any of the studied concentrations. The experimental results demonstrate a simple, rapid and affordable UV-Vis spectrophotometric method that could be applied to the quantitation of TRI in this ternary complex.


A triancinolona (TRI) é um fármaco amplamente utilizado no tratamento de doenças inflamatórias do globo ocular e é praticamente insolúvel em água, o que limita sua utilização na forma de colírio. As ciclodextrinas (CDs) têm sido utilizadas com sucesso para aumentar a solubilidade ou velocidade de dissolução de fármacos. O presente estudo teve como objetivo a validação de uma metodologia analítica para a TRI a partir de complexos de inclusão com beta-ciclodextrina (B-CD) associada com a trietanolamina (TEA) (complexo ternário) por espectrofotometria de UV-Vis. A validação do método proposto foi realizada de acordo com os parâmetros analíticos estabelecidos pela Agência Nacional de Vigilância Sanitária (ANVISA). As análises quantitativas foram realizadas a 242nm a temperatura ambiente, utilizando cubeta de quartzo de 1cm. Os estudos de precisão e exatidão foram realizados para cinco níveis de concentração (4, 8, 12, 18 e 20µg.mL-1). A B-CD associada a TEA não alterou o comportamento fotoquímico da TRI. Os resultados da avaliação dos parâmetros analíticos demonstraram o sucesso da metodologia. A curva padrão apresentou linearidade (r2 > 0.999) na faixa de concentração de 2 a 24µg.mL-1. A metodologia apresentou bons níveis de precisão para o estudo inter dia (desvio padrão relativo-DPR <3.4%) e reprodutibilidade (DPR<3.8%). A exatidão ficou em torno de 80% e a variação de pH inserida no estudo de robustez não revelou uma interferência significativa em todas as concentrações estudadas. Os resultados experimentais demonstraram um simples, rápido e viável método de espectrofotometria de UV-Vis com aplicabilidade para a análise quantitativa da TRI a partir do complexo ternário.
